Can't think of any non-animated good comedy series running right now. We're always a couple of months behind with TV series, anyway.

Two and a Half Men is pretty decent, but I thought it became quite boring after a season or two.
I still like Frasier, Becker etc. They broadcast both shows daily on Comedy Central. I've seen about every single show by now, but the huge amount of Frasier seasons at least makes the first episodes feel kind off fresh when they start all over again.
Still can't beat the current animated sitcoms, but still.
Funniest show I can think of, although not being a sitcom, is The Daily Show with Jon Stuart. It doesn't air too often here (As we only get the dumbed down global edition) and, again, gets a pretty crappy timeslot, but it might just be the funniest show on TV.
